用大孔树脂来分离和纯化苯乳酸

论文摘要

苯乳酸由乳酸菌生产,它是一种重要的保鲜剂。苯乳酸是一个重要的防腐剂以及可以延长食品的保质期。本论文对苯乳酸进行研究其分离和纯化方法,并且研究它跟树脂的静态吸附和动态吸附。6种树脂(301,315,335,345,HZD-9,HZ-2)用来考查它们的吸附量和解吸量。结果表示301树脂的效果最好,因此以它进行下面的研究。首先将发酵液在活性炭加热处理(温度为80℃),活性炭用量为5%。然后将发酵液的苯乳酸吸附在301树脂再用盐酸来洗脱被吸附的苯乳酸;萃取之前将流出溶液进行pH调解再用乙酸乙酯萃取。最后,用旋转蒸发器浓缩萃取溶液使苯乳酸结晶。HPLC流动相为A(三氟乙酸0.05%/水)/B(三氟乙酸0.05%/甲醇)50:50(v/v),流速为1ml/min。苯乳酸保留时间在5分钟左右。301树脂对苯乳酸的吸附过程比较符合Langmuir方程,关系系数R2为0.99,自由能(△G)为负值。吸附过程的解析率和产品的纯度分别为87.31%和95.07%。
