论文摘要
目的研究血管生成抑制剂Endostatin（内皮抑素）、SU6668联合5-Fu对裸鼠结肠癌移植瘤生长和转移的抑制作用。方法建立人结肠癌裸鼠原位种植转移模型。将60只荷瘤鼠随机分为5个组,每组12只。种植12天后分别自腹腔内注射生理盐水（对照组）、Endostatin（E治疗组）、SU6668（S治疗组）、Endostatin + SU6668（E+S治疗组）、Endostatin + SU6668 + 5-Fu（E+S+F治疗组）,每天1次,共用28天。种植后第42天采用颈椎脱位法处死裸鼠,观察肿瘤腹膜和区域淋巴结转移及腹水出现情况;秤取原位肿瘤的瘤重、计算抑瘤率;采用免疫组织化学染色并测定肿瘤微血管密度（microvessel dentisy, MVD）。结果对照组、E治疗组、S治疗组、E+S治疗组、E+S+F治疗组瘤重（g）分别为（1.36±0.07）、（0.70±0.08）、（0.75±0.07）、（0.55±0.05）、（0.45±0.05）;抑瘤率（%）分别为0、48.5、44.9、59.6、66.9;腹膜转移率（%）分别为70、8.3、18.2、0、0;区域淋巴结转移率（%）分别为90、16.7、18.2、0、0;腹水出现率（%）分别为40、0、0、0、0。MVD分别为（13.0±1.33）、（4.00±0.40）、（4.10±0.32）、（2.19±0.26）、（1.38±0.19）; E治疗组、S治疗组、E+S治疗组、E+S+F治疗组与对照组相比,结肠癌生长和转移受到明显抑制,尤以E+S+F治疗组最显著。结论Endostatin、SU6668联合5-Fu对结肠癌移植瘤的生长和转移有强烈的抑制作用,三者具有协同效应。
