脑源性神经营养因子对急性精神应激的保护作用和氟西汀的干预研究

目的:研究精神应激后部分脑区内BDNF、CREB蛋白及mRNA表达的时序性变化,进一步探讨BDNF对精神应激的保护作用及其机制。 方法:采用旁观电击大鼠应激模型作为精神应激模型,以旷场实验、高架十字迷宫实验评估大鼠的焦虑行为,利用免疫组织化学、原位杂交技术,观察精神应激后海马CA1区、CA3区、齿状回、前额叶皮质、杏仁核中央核、伏隔核(壳区)、中脑导水管周围灰质、腹侧背盖区、下丘脑背内侧核等脑区内BDNF、CREB蛋白及mRNA不同时点的表达。 结果: 1.行为学评定结果:在旷场实验中,外周格数、中央格数、以及修饰数均是生理应激组低于精神应激组,精神应激组低于正常对照组,差异具有显著性;在高架十字迷宫实验中,开放臂次数以及开放臂时间也是生理应激组最低,精神应激组居中,正常对照组最高,差异具有显著性。 2.精神应激后即刻,以上各脑区内BDNF蛋白及mRNA表达均明显下调,灰度值明显高于对照组,差异具有极显著性;精神应激后0.5h、1h BDNF蛋白及mRNA表达开始恢复,但仍少于对照组,差
