作者杨瑞成(2019)在《致脑膜炎大肠杆菌诱导EGR-1介导血脑屏障通透性破坏的分子机制研究》一文中研究指出:细菌性脑膜炎对于人类健康和畜禽养殖都是一个巨大的挑战。在人医方面细菌性脑膜炎为十大引发高死亡率的感染性疾病之一,并且大部分的幸存者会留下不同程度的神经系统后遗症;在兽医方面大量养殖动物由于患脑膜炎失去了其经济价值,给畜牧产业造成了巨大的经济损失。肠外致病性大肠杆菌(Extraintestinal pathogenic Escherichia coli,ExPEC)近年来受到了广泛的关注,它可以感染宿主多个器官,导致尿路感染以及脑膜炎等。同时,ExPEC还能感染宠物导致疾病的交互传播,污染食物造成食品卫生安全恐慌。因此,ExPEC引起不同宿主的相关疫病已经成为严重影响人类健康、食品安全和畜牧养殖业发展的一类重要人畜共患传染病。目前为止,关于ExPEC脑膜炎的研究主要集中于E.coli侵入中枢神经系统(Central nervous system,CNS)过程中侵入相关宿主靶基因的筛选。而对于,参与致脑膜炎E.coli破坏血脑屏障(Blood-brain barrier,BBB)通透性过程的宿主基因的功能报道较少。本研究以临床分离的不同种属来源的ExPEC为基础,结合致脑膜炎E.coli感染人脑微血管内皮细胞(Human brain microvascular endothelial cells,hBMECs)差异mRNAs和非编码RNAs测序结果,探索致脑膜炎E.coli诱导中枢炎症过程中介导BBB通透性增加的分子机制。在本研究中,将本实验室保存的7株人源、20株猪源和3株禽源ExPEC菌株分别进行了细菌侵入hBMECs细胞以及小鼠脑组织定植实验。共鉴定发现了PCN033、Xiantao10、ACN005、PCN079、HCN10618、PCN080、A764以及B958等8株侵入hBMECs细胞能力较强的菌株,其中PCN033和A764这两株菌株在小鼠血液中的存活能力以及脑组织中的定植能力较强。由于PCN033分离自发病猪的脑脊液,所以选择PCN033作为后续实验菌株。同时,我们发现致脑膜炎E.coli通过下调紧密连接蛋白(Tight junctions,TJs)在hBMECs细胞上的表达以及改变其在细胞上的分布情况增加BBB的通透性。随后,利用全转录组测序技术对致脑膜炎E.coli感染hBMECs细胞胞内差异表达的mRNAs和非编码RNAs进行了分析,共鉴定到366个mRNAs、895个lncRNAs(Long non-coding RNAs)、32个miRNAs(Micro RNAs)以及308个circRNAs(Circular RNAs)发生了差异显著的变化,并绘制了差异变化的circRNAs、miRNAs以及mRNAs的调控网络关系图。通过分析致脑膜炎E.coli感染hBMECs细胞全转录组数据,我们挖掘了致脑膜炎E.coli感染宿主破坏BBB完整性过程中的核心宿主因子EGR-1的生物学功能。通过hBMECs EGR-1基因缺失细胞系和EGR-1基因缺失小鼠,分别从体内和体外证明了EGR-1对于中枢炎症以及BBB通透性的调控作用。我们发现转录调节因子EGR-1在感染早期显著上调表达,可以入核直接调控VEGFA、PDGFB、ICAM-1以及TNFα的表达。通过更进一步的体内、体外实验,我们发现致脑膜炎E.coli感染宿主上调EGR-1的表达诱导VEGFA和PDGF-BB大量分泌,血液循环中过量存在的VEGFA和PDGF-BB作用于BMECs细胞导致TJs表达量下降且分布发生改变。在另一方面,致脑膜炎E.coli感染宿主诱导转录调节因子Snail-1显著上调,Snail-1可以直接入核负调控ZO-1和Occludin等TJs的表达,同样起到促进BBB通透性增加的作用。同时,致脑膜炎E.coli感染宿主上调EGR-1的表达诱导BMECs细胞活化指示蛋白ICAM-1显著上调,在感染初期ICAM-1在BMECs细胞上的大量表达直接促进免疫细胞穿越BBB进入脑实质介导CNS炎症反应,进而造成BBB通透性增加。此外,本研究首次在细菌性脑膜炎模型中,对circRNAs的功能进行了研究,发现致脑膜炎E.coli感染hBMECs细胞之后hsacirc2858的表达显著上调,并且可以通过海绵作用吸附hsa-miR-93-5p介导VEGFA的上调表达。本研究阐明了EGR-1在致脑膜炎E.coli感染宿主导致BBB通透性增加以及神经炎症过程中的功能,为今后细菌性脑膜炎的预防和治疗提供了新的思路和潜在靶点。
